I'm a bit new to domaining so far, and just want to make sure on this. When you're looking for how many times keywords show up in Google, do you search with or without quotes?

I would imagine you're searching with quotes? For example, when searching for unreal tournament forums, Google would give 6.9 million results, but only 14.000 when searching for the same keywords in quotes -> "unreal tournament forums".

I would imagine you take into consideration the results for the search with quotes right?

With quotes is the best way.
For instance if the domain was FishTacos.com, the best way to search is "Fish Tacos" since that gives an exact keyword match.

I agree, if you do not use quotes, then google will bring up the words you are searching or, whether they are searched for together or individually. Using quotes, google will only bring up results that have the two words searched together!.

I would suggest to get a feel of the competition that is going on for your keywords, do a search like this:

intitle:"fish tacos"

That will display the number of pages out there that holds your keywords in the page title, a high number of results means a lot of people are optimizing for it. (SEO).

That would give you a better idea in my opinion of the amount of possible end users, thus the possibility your domain could sell.

Depending of course how far you're going with the amount of keywords in the domain, 2 - 3 keywords is the limit i would use.
And the type of keywords...

Natural sounding keyword combinations versus combinations that makes no sense, what niche?

You would want to have natural sounding keyword combinations and a niche where there is monetary value.
