Does a domain with 40 Overture result with extension necessarily have a traffic?

No, not necessarily, but if it's a good name (nice name, ext...) then probably it's a good bet.

Don't see only the big! I had one with 26 and was getting 100 visits per month and 1 with 30 and was getting only 20 visits per month

If it is a nice name with .com extension I'd reg it.

There are many variables involved to safely say for sure without knowing the name.

If there's a hyphen involved, then probably not.
If it's a typo (either generic or of an established website) most likely yes.
If it's a combo word that you're not sure about, make sure there are results with the words together and separated.
If you can't figure out why the name would be getting traffic, try the other extentions - foreign as well - as it very well be traffic leaks from an established foreign site. (i.e. Germans typing in the .com version of a .de site and vice versa)
Look up the term in google to see if there's a reason for the term being in existence.

That's all I can think of right now.

also try plural variation and see if there is active site there. traffic can come from there.

Overture is a pay per click search engine and it provides a keyword tool to help it's advertisers select the right keywords when they buy ads.

You enter a keyword(s) and it will show you the number of searches made the previous month by surfers using overture's network of sites.

Domainers use this tool (among others) to find key phrases and register domains. Usually, when you enter a keyword in the tool followed by a tld (i.e. "cameras.com") if you get any results, chances are the particular domain gets some traffic.

Go play with it to get a feel with how it works. It can be tricky and the results do not always reflect the truth, so don't go registering domains right away because most likely you'll get burned.

Elsewhere in this forum (and others) people occasionally post helpfull info about the tool, so I suggest you read these to get yourself better acquainted with how it works.
