dev.mobi recommends developing .mobi sites for a maximum width of 200 pixels to hit the sweet spot of mobile devices. However, this will probably change quickly due to advancements in technology.

As far as when browsing using Explorer or Firefox, the best situation is to have sites auto-detect that the user is on a PC and render a full-screen webpage (IMO...).
