Optimizing in photoshop

I am experimenting with building a full site in Photoshop to take advantage of some of the graphical tricks in the program.

Can anyone tell me how to optimize the graphics in Photoshop or leave certain areas out when I save as a web page so that I can put my own graphics in and/or reduce my file size?

you mean making images websafe or transparent?
