I don't think so.
You don't need to know JS in order to develop an AJAX application. Microsoft is developing an tech call Atlas for Ajax development in ASP.NET 2.0. Let the framework do the hard work.

GMail is a good example of AJAX. Your web application performs just like a desktop application. Actually AJAX is a client-side script.

When I do AJAX, it is basicly a short JS script which is the basis of all AJAX and the rest is PHP.

Since AJAX (Asynchronous Javascript And XML) is just a combination of already excisting standards ((X)HTML, JavaScript, XML) it depends on your personal skills. If you're a experienced webdesigner, it doesn't take much effort to pick it up.

I only apply it in certain web apps, for some it totally works and it's definitely the way to go, for others there are better solutions. Keep in mind that AJAX gives you trouble with some older browser versions.

I tend to veer away from AJAX due to the fact I have heard rumors it does not work with all browsers.
